preparation method | β-alanine (β-Ala)(0.5g,6.61mmol) was dissolved in 10% NaCO3(12ml) under stirring in a 50ml flask. Gradually add Fmoc-Cl(1.45g,5.61mmol) of dioxane (10ml) solution to the resulting solution that has been placed in an ice bath. The reaction mixture was stirred at room temperature for 4 hours, then water (80ml) was added. The effluent phase layer was separated from the reaction mixture and stripping with ether three times (75ml × 3). The stripping water layer was acidified with an aqueous solution of 2NHCl to a pH of 2, and then extracted with ethyl acetate three times (75ml × 3). The organic phase layer was recovered and concentrated to obtain 1.50g of crude product. The crude product was recrystallized in a mixed solvent (30ml) of ethyl acetate: hexane = 1 ∶ 2, and filtered under reduced pressure to obtain a white solid, expressed as FMOC-beta-alanine. |
